麦长管蚜唾液中几种酶的鉴定、活力测定与功能分析   总被引:1,自引:1,他引:0  
用Parafilm膜夹营养液法,以两种食料介质饲喂麦长管蚜Macrosiphum avenae 3龄若蚜并收集其唾液,对唾液中的酶类进行了鉴定、活力测定和功能分析。结果表明,在20%蔗糖介质提取液中,鉴定有果胶酶、多酚氧化酶和纤维素酶; 在水介质提取液中鉴定有纤维素酶; 两种介质提取液中都未鉴定出过氧化物酶。酶活力测定结果表明, 在20%蔗糖介质提取液中, 每30头蚜虫分泌的果胶酶、多酚氧化酶和纤维素酶的酶活力分别为2.59×10-3 U/g、7×10-3 U/g和7.89×10-3 U/g; 在水介质提取液中,纤维素酶活力为3.68×10-3 U/g。行为反应试验结果表明,果胶酶处理麦苗的挥发物组分能引起麦长管蚜寄生性天敌燕麦蚜茧蜂Aphidius avenae和捕食性天敌七星瓢虫Coccinella septempunctata 的嗅觉偏好反应,因此,果胶酶在麦长管蚜取食诱导小麦植株的间接防御反应中具有重要作用。  相似文献

刺吸式昆虫在刺吸作物韧皮部取食后会影响作物的正常生长发育,随着作物抗性及昆虫种类的不同,作物反过来也会对昆虫造成或正或反的影响,然而一种刺吸式昆虫取食后对后来者有什么样的影响目前尚不确定。本研究通过严谨的实验方法,即选定3个不同国家的小麦抗性品种98-10-30、Amigo和Batis,在所有条件严格统一的人工智能控制温室内,通过各处理在同一植株上先后不同的接蚜方式,分别测定了麦二叉蚜Schizaphis graminum (Rondani)和麦长管蚜Sitobion avenae Fab.在前期被蚜虫危害的抗虫品种98-10-30、Amigo和Batis上的发育历期、体重差及相对日均体重增长率(mean relative growth rate MRGR)等生物学参数。结果表明:品种不同,蚜虫在不同处理条件下取食作物时受到的影响也不同,即在品种98-10-30上,通过与前期不接虫的对照及前期接不同麦蚜的相关处理比较,麦长管蚜对后期取食的麦二叉蚜(MRGR=0.0974±0.0071)具有抑制作用,而麦二叉蚜对后期取食的麦长管蚜(MRGR=0.1614±0.0048)却有促进作用;在品种Amigo上,前期麦蚜的危害对麦长管蚜的取食具有促进作用,而在品种Batis上前期危害对麦二叉蚜的取食具有促进作用。同时明确了3个品种对两种蚜虫的抗性状况,即在前期无蚜虫危害时,品种98-10-30和Batis对两种麦蚜的抗性相当;在前期有麦蚜危害时,品种98-10-30对麦二叉蚜的抗性较好,而品种Batis对麦长管蚜的抗性较好;品种Amigo无论在任何处理下均对麦二叉蚜的抗性较好。  相似文献

Elevated concentrations of atmospheric CO2 can alter plant secondary metabolites,which play important roles in the interactions among plants,herbivorous insects and natural enemies.However,few studies have examined the cascading effects of host plant secondary metabolites on tri-trophic interactions under elevated CO2(eCO2).In this study,we determined the effects of eCO2 on the growth and foliar phenolics of Medicago truncatula and the cascading effects on two color genotypes oiAcyrthosiphon pisum(pink vs.green)and their parasitoid Aphidius avenae in the field open-top chambers.Our results showed that eCO2 increased photosynthetic rate,nodule number,yield and the total phenolic content of M.truncatula.eCO2 had contrasting effects on two genotypes of A.pisum;the green genotype demonstrated increased population abundance,fecundity,growth and feeding efficiency,while the pink genotype showed decreased fitness and these were closely associated with the foliar genstein content.Furthermore,eCO2 decreased the parasitic rate of A.avenae independent of aphid genotypes.eCO2 prolonged the emergence time and reduced the emergence rate and percentage of females when associated with the green genotype,but little difference,except for increased percentage of females,was observed in A.avenae under eCO2 when associated with the pink genotype,indicating that parasitoids can perceive and discriminate the qualities of aphid hosts.We concluded that eCO2 altered plant phenolics and thus the performance of aphids and parasitoids.Our results indicate that plant phenolics vary by different abiotic and biotic stimuli and could potentially deliver the cascading effects of eCO2 to the higher trophic levels.Our results also suggest that the green genotype is expected to perform better in future eCO2 because of decreased plant resistance after its infestation and decreased parasitic rate.  相似文献

During the years 1989–1992 cereal aphids were caught alive in a low level (1.5 m high) suction trap operated in Le Rheu (Brittany, France) and tested for BYDV transmission. In most cases comparisons with data collected simultaneously by a 12.2 m suction trap operating in the same site resulted in good relationships between weekly catches at both heights. Results from transmission tests showed that: (i) the two main BYDV vectors were Rhopalosiphum padi and Metopolophium dirhodum during the years of experiment; (ii) PAV and MAV were the commonest viruses and RPV was relatively scarce; (iii) during spring M. dirhodum appeared to be the most important MAV vector and nearly as good a PAV vector as R. padi; (iv) during autumn R. padi was the only vector of the three viruses with mixed transmission allowing it to transmit also MAV probably by heteroencapsidation. To give an indication of the risk of infection, infectivity indices were calculated by multiplying the numbers of aphids caught by the 12.2 m suction trap by the proportion that were infective. Extended sieve element salivation (E1 waveform in the electrical penetration graph) is a characteristic activity during early sieve element punctures, particularly in resistant plants. In order to explore a chemically-mediated mechanism of resistance associated with sieve element salivation, we compared the pattern of feeding behaviour of the aphid, Sitobion fragariae (Walker), on two cultivars of the wheat Triticum aestivum L., with different concentrations of hydroxamic acids (Hx). During 24 h of electronic monitoring, aphids dedicated over 50% of the total time to phloem ingestion from the sieve elements. Total time allocated to E1 in the experiment, time to first E1 within the experiment, time allocated to E1 before a sustained phloem ingestion (E2) and the contribution of sieve element salivation to the phloem phase (E1/[E1+E2]) were significantly higher in the high-Hx cultivar. The increased salivation in plants with higher contents of Hx suggests the existence, at least in this system, of a chemically-mediated sieve element constraint.  相似文献

Peroxidases (POXs) make up a large superfamily of enzymes that act in a wide range of biological mechanisms, including maintaining appropriate redox balances within cells, among other actions. In this study, we cloned a sequence that encodes a POX protein, SaPOX, from wheat aphids, Sitobion avenae. Amino acid sequence alignment showed the SaPOX sequence was conserved with POXs from other insect species. SaPOX mRNA accumulations were present in all nymphal and adult stages, at higher levels during the first and second instar, and lower during later stages in the life cycle. Ingestion of dsRNA specific to POX led to reduced SaPOX mRNA accumulation. Sitobion avenae nymphs continuously exposed to dietary dsPOX via an artificial diet led to reduced survival rate and ecdysis index. We infer that POX is important to maintain the growth and development of S. avenae.  相似文献

Entomophthoralean-specific primers were used to estimate the occurrence of entomophthoralean species in the cereal aphid Sitobion avenae in Denmark. Sequencing documented for the first time the presence of Pandora kondoiensis in Denmark. Many insects harbour facultative endosymbiotic bacteria, often more than one type at a time. These symbionts can have major effects on their hosts' biology, which may be modulated by the presence of other symbiont species and by the host's genetic background. We investigated these effects by transferring two sets of facultative endosymbionts (one Hamiltonella and Rickettsia, the other Hamiltonella and Spiroplasma) from naturally double‐infected pea aphid hosts into five novel host genotypes of two aphid species. The symbionts were transferred either together or separately. We then measured aphid fecundity and susceptibility to an entomopathogenic fungus. The pathogen‐protective phenotype conferred by the symbionts Rickettsia and Spiroplasma varied among host genotypes, but was not influenced by co‐infection with Hamiltonella. Fecundity varied across single and double infections and between symbiont types, aphid genotypes and species. Some host genotypes benefit from harbouring more than one symbiont type.  相似文献

为了探索高温防控麦蚜的技术,本实验设置系列短时间高温处理(45℃,30、40、50、60、70、80、90 s;50℃,5、10、15、20、25、30、35、40 s;55℃,5、10、15、20 s;60℃,5、10、15 s;65℃,5、10 s;70℃,5 s),测定了麦长管蚜Sitobion avenae(Fabricius)若蚜及成蚜的死亡率。结果表明:在同一温度下,随着处理时间的延长,麦长管蚜的死亡率上升;在相同处理时间下,随着温度的升高,其死亡率上升。在45℃条件下,需要近90 s其死亡率才能达到100%;在70℃下,100%死亡率仅需5 s。麦蚜的半致死温度随着处理时间的延长而降低,麦蚜的半致死时间随着处理温度的上升而缩短。通过logistics模型分析发现,在相同处理下,若蚜的半致死温度和半致死时间均高于成蚜。  相似文献
